Acetylcholine (Ach) is a major excitatory neurotransmitter that regulates the movement of esophageal smooth muscle. The contraction of esophageal smooth muscle mainly results from Ach release. The choline acetyltransferase (ChAT) and acetylcholinesterase (ACHE) are synthase and hydrolase of Ach, respectively, to regulate the metabolism of Ach. The activities of the two enzymes in the esophageal tissue represent the endogenous cholinergic function of the esophagus. Acetylcholine is the major neurotransmitter of the spinal cord motor neurons, autonomic nervous system ganglion fibers, postganglionic cholinergic (parasympathetic) fibers, and plenty of neurons in the central nervous system (such as the basal ganglia, the motor cortex of the brain). Acetylcholine is synthesized by the reaction of choline and mitochondrial acetyl-CoA under the catalysis of ChAT. After release, acetylcholine stimulates specific cholinergic receptors. The local cholinesterase will quickly terminate the interaction and hydrolyze acetylcholine into choline and acetate. Acetylcholine levels are regulated by the intake of ChAT and choline.||The Choline Acetyltransferase (ChAT) BioAssay™ Kit can be used to measure Choline Acetyltranferase (ChAT) activity in tissue samples. ChAT is synthesized in the perinuclear body of neurons and transported to neurons through the mechanism of slow axon transport. ChAT is the most specific indicator for monitoring the functional status of cholinergic neurons in central and peripheral nervous systems.||Test Principle:|Acetyl-CoA and choline react under the catalysis of Choline acetyltransferase (ChAT) to produce coenzyme A (CoA), CoA can combine with the 4, 4-dithiopyridine.
