This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal IP3. During the reaction, Universal IP3 in the sample or standard competes with a fixed amount of Universal IP3 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal IP3. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal IP3 in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Inositol-1, 4-bisphosphate (Ins 1-(1, 4)P2) and inositol-1, 4, 5-triphosphosate (IP3). They are derived in the body from enzymatic hydrolysis of phosphatidyl inositol substances (e.g., phospholipase C). Its main effect is to induce the release of Ca2+ from the intracellular reservoir and instantly increase the concentration of Ca2+ in the cellular fluid. This process is based on the fact that Ca2+ is a widespread intracellular messenger, so it plays an important regulatory role in cellular response, and it is complementary to the regulatory effect of diacylglycerol.
