-20°CRecognizes a polypeptide which is identified as insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as beta-cell and insulinoma marker.||Applications: |Suitable for use in Immunofluorescence, ELISA, Flow Cytometry (Not Tested), Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Flow Cytometry (Not Tested): 0.5-1ug/million cells |Immunofluorescence: 1-2ug/ml |Western Blot: 0.5-1ug/ml |Immunoprecipitation: 1-2ug/500ug protein lysate|Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1ug/ml for 30 minutes at RT|(No special pretreatment is required for staining of formalin/paraffin tissues.|Optimal dilutions to be determined by the researcher.||Positive Control:|Pancreas||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. |Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.||Note: Applications are based on unconjugated antibody.
