-20°CRecognizes a conserved determinant of the p53 protein. Its epitope maps near the C-terminus end (aa 370-378) of p53. It has been used successfully to immunoprecipitate p53 oncoprotein complexes with simian virus 40 (SV40) large T antigen. Reportedly, this antibody precipitated the most ATPase-active form of T. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.||Applications: |Suitable for use in Immunofluorescence, ELISA, Flow Cytometry (Not Tested), Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution: |Flow Cytometry (Not Tested): 0.5-1ug/million cells |Immunofluorescence: 0.5-1ug/ml |Western Blot: 0.5-1.0ug/ml |Immunoprecipitation: 0.5-1ug/500ug protein lysate|Immunohistochemistry: Frozen only: 0.5-1.0ug/ml for 30 minutes at RT |Optimal dilutions to be determined by the researcher.||Positive Control:|MDA-MB-231 Cells. Breast or Colon carcinoma||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. |Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.||Note: Applications are based on unconjugated antibody.
